Description

CUR Series scalable bioreactor platform

Developed independently by JYSS BIO, the CUR Series Torrent Bioreactor is a single-use bioreactor that uses a non-bubbling interfacial oxygen-transfer mechanism for mixing. While meeting large-volume production requirements, this advanced mass-transfer method helps avoid cell damage caused by bubble surface tension and the high shear generated by impellers in conventional bioreactors.

Within the normal operating speed range, the mean shear rate inside the reactor can be lower than 20 s-1, enabling high-density, high-viability culture of shear-sensitive cells. The series provides working volumes from 0.5 L to 2000 L for cell therapy, gene therapy, monoclonal antibody and vaccine applications.

Operating Principle

Stable wave motion and interfacial oxygen transfer across scales

The vessel moves in an overall planar circular motion, driving the liquid to form waves that wash the side walls and transfer oxygen into the liquid phase. This enables efficient oxygen dissolution in an extremely low-shear environment. The resulting stable and gentle flow field supports the growth and proliferation of fragile cells.

Applications and Cell Culture

Suitable for suspension culture of HEK293, CHO, T cells, NK cells, iPSC and HS cells. Supported processes include batch, fed-batch and perfusion culture.

CHO Cell Perfusion Culture

During CHO cell perfusion culture in a CUR50 reactor, cell viability remained above 98%. The product literature reports a final protein expression level approximately 20% higher than with the compared culture methods.

HEK293T Cell Culture

For lentivirus production, cell density increased from 1.0×106 cells/mL to 6.0×106 cells/mL within three days, while viability remained above 90%. Viability stayed at 90% after transfection.

Monoclonal Antibody Culture and Scale-Up

CUR5L, CUR50L and CUR500L showed similar process performance, stability and scale-up consistency. Under the reported comparison conditions, peak cell density was at least 50% higher and monoclonal antibody yield was 40% higher than in the compared JYSS BIO agitator tanks using the same process.

System Components and Control

The system comprises a control tower and tank assembly. The tank consists of an oscillator and culture vessel. Outer surfaces use brushed 304 stainless-steel plates in line with GMP requirements, and electrical parts are CE certified.

Control Tower

Remote monitoring; real-time acquisition of pH, DO, temperature and speed; report and trend generation; Ethernet and USB interfaces; optional weighing, jacket temperature-control and perfusion modules.

Dissolved Oxygen Mixing System

Planar eccentric motion and a single-axis drive promote thorough gas-liquid mixing with low shear and high oxygen-transfer efficiency.

Inverted Truncated-Conical Vessel

The cylindrical-wall and conical structure creates a relatively large gas-liquid interface, supporting oxygen transfer and CO2 removal.

Online Measurement

The pH module can link acid and alkali pumps with CO2. The DO module can link N2, O2 and air valves for closed-loop control and gas-flow recording. Manual HMI control is also available.

Temperature and Pumping

A heating film provides continuous heating. An optional jacket module supports heating and cooling. Bidirectional peristaltic pumps can be used for medium addition, harvest, acid and alkali addition, and perfusion.
